Il film tratterà le vicende che chiuderanno la carriera liceale di Troy (Zac Efron), Gabriella (Vanessa Hudgens), Sharpay (Ashley Tisdale) e Ryan Evans (Lucas Grabeel) e degli altri personaggi. Durante le preparazione della festa di fine anno, prepareranno uno spettacolo musicale, il quale parlerà delle loro esperienze e del loro futuro.
E' stato confermato che ci saranno 14 numeri musicali nel film, e che quindi la colonna sonora sarà composta da 14 brani. Inoltre la colonna sonora includerà una canzone apposita per i crediti finali che vedrà protagonista il vincitore di "High School Musical: Get in the Picture".
